    Yes, that makes sense.

    Normally, not so in your case thus far, when importing one column == one custom field, usually named to the same as the column title in the spreadsheet.

    Doing this makes the data more searchable.

    You might want to make use of the data twice. Once as you have, into the post contents nicely formatted, and then individually into the custom fields. Otherwise, if you don’t put it also in the content, you then have some custom coding to do to get the custom fields to output.

    You need to check if the import plugin that you are using can import into ACF Custom fields (it might require the Pro version of the import plugin).

    CSV should work fine, but ALWAYS take a database backup before you do anything!!! There are plenty of free plugins that will do this for you.
